AOD-9604: The Fat Loss Fragment of GH

📅 July 23, 2026 ⏱️ 2 min read ✍️ Team TPS

AOD-9604 is the fat loss fragment of growth hormone with the growth removed. That is the entire pitch, and whether it works is a more complicated question.

What it is

AOD-9604 reproduces amino acids 176-191 of human growth hormone, the C-terminal fragment associated with fat metabolism.

The theory is elegant: isolate the region responsible for lipolysis and you get the fat loss effect without the growth-promoting activity, the insulin resistance, or the tissue growth that full GH brings.

Why that is appealing

Full growth hormone raises blood glucose, can promote growth in tissues you did not intend, and is expensive. A fragment with none of those properties would be genuinely useful.

AOD-9604 does appear to lack GH’s effect on IGF-1 and blood sugar, which supports the basic premise.

The efficacy problem

Early animal work was promising. Human trials were less so, the clinical development programme did not produce the weight loss results needed to proceed, and the compound was not approved for obesity.

That is the crux. AOD-9604 is well tolerated and appears safe; what it has not convincingly demonstrated is that it works well enough in humans to matter.

How it compares to what does work

The GLP-1 compounds produce large, well-documented weight loss through appetite reduction. Comparing AOD-9604 to tirzepatide or retatrutide is not close, and anyone choosing a fat loss compound on evidence should know that.

Where AOD-9604 retains interest is the safety profile: no appetite suppression, no gastrointestinal effects, no hormonal suppression. For someone who cannot tolerate a GLP-1, that is not nothing.

The joint claim

More recent interest has focused on cartilage and osteoarthritis rather than fat loss, which may prove the more durable application.

Where it fits

A low-risk, low-expectation option. Not a serious fat loss strategy on current evidence, and not a substitute for a deficit. See GLP-1 side effects explained for the compounds that do move the needle.
